This study aims to evaluate Sudanese Hibiscus sabdariffa (Roselle) extract as an alternative counterstain to Eosin in histopathology. The study is based on the need to explore natural, locally available, cost-effective, and environmentally friendly staining alternatives for use in medical laboratories. Hibiscus sabdariffa extract will be prepared from dried plant material and used as a counterstain in histological sections instead of Eosin. The staining performance will be compared with the conventional Hematoxylin and Eosin (H&E) staining method based on staining intensity, cytoplasmic clarity, contrast, cellular detail, and background staining. The study is expected to determine whether Sudanese Hibiscus sabdariffa extract can provide acceptable staining quality comparable to Eosin. The findings may support its potential use as a natural and locally available alternative counterstain in histopathology laboratories, while promoting the utilization of Sudanese plant resources and reducing the cost and environmental impact of conventional staining materials.

Participant confidentiality will be maintained by not recording participants’ names or other identifying information on research forms or specimen slides. Each specimen will be assigned a unique study code instead of the participant’s name. All collected data and specimens will be securely stored and accessible only to the research team. The information and specimens will be used for research purposes only and will not be disclosed to unauthorized persons.
This study involves minimal risk to participants, as no additional invasive procedures or interventions will be performed. The study will use tissue specimens already collected for routine diagnostic purposes. The main potential risk is a possible breach of confidentiality, which will be minimized by coding all specimens and protecting participants’ personal information
The study may not provide direct benefits to individual participants. However, it may contribute to the development of a natural, locally available, cost-effective, and environmentally friendly alternative counterstain to Eosin in histopathology laboratories. The findings may also promote the utilization of Sudanese Hibiscus sabdariffa in scientific research and laboratory practice.
